**Day Grab** samples were collected at 76 sites within Coquet to St Marys Marine Conservation Zone on survey CEND0716 The 0.1 m2Day grab was used throughout to collect sediment samples for infaunal community analysis, PSA and sediment contaminant samples. A single grab sample was collected from within a 50 m radius around each target location for PSA and macrofaunal analysis. On recovery of each grab sample, a photograph of the sample was taken in situ and the sample depth measured to ensure the minimum sample size of 5 cm was obtained. A core of approximately 5 cm was collected for PSA before the remaining sample was processed for macrofaunal analysis. The macrofaunal sample was decanted into a plastic box and transferred to the sample processing area on board the vessel. Benthic fauna were collected by washing the sample with sea-water over a 5mm and 1 mm sieve. The retained >1 mm fraction was transferred to a labelled container and preserved in buffered 4% formaldehyde for later analysis ashore. A visual assessment was made of the sediment type sampled by the grab and noted on the field records, assigning the sample to a Folk class and its equivalent EUNIS and BSH sediment classes. **NIOZ Core** samples were collected at 45 sites within Coquet to St Marys Marine Conservation Zone on survey CEND0716 The 0.1 m2 NIOZ standard box corer was used to collect undisturbed sediment samples of up to 50 cm in depth at selected stations. A minimum of three replicate cores spaced a minimum of 5m apart were collected from within a 50 m radius around each target location. On recovery of the NIOZ corer, a photograph of the sediment surface was taken in situ. A 5 cm diameter (30 cm long) sub-corer was used to obtain a PSA sample which was sliced at 5 cm intervals and placed in separate plastic bags for analysis. The remaining sample was then carefully excavated from the core tube for faunal evidence collection to verify the mud feature within Coquet to St Mary’s MCZ: Survey Report 14 analysis. Faunal analysis was performed on the sediment which had been carefully extracted from the core tube following the same procedure that was carried out for Day grab sampling with the retained >1mm fraction placed into two labelled containers and preserved in buffered 4% formaldehyde for later analysis ashore. NIOZ standard box corer (0.1m2)
